Report for Colostrum Powder Samples

Introduction:

Xenobiotic Detection Systems, Inc. (XDS) has a genetically engineered cell line which contains the firefly luciferase gene under trans-activational control of the aryl hydrocarbon receptor (patent # 5,854,010).  This cell line can be used for the detection and relative quantitation of AhR agonists and for identification and characterization of novel AhR agonists. Our term for the in vitro assay is Chemical-Activated Luciferase Expression or CALUX™ assay. The most widely studied class of compounds that activate this system is the polychlorinated diaromatic hydrocarbons (PCDH), such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD, dioxin). Many PCDH compounds are quantitated versus relative activity to TCDD, since this is one of the most potent activators of AhR-mediated gene transcription. This relative quantitation is known as Toxic Equivalents (TEQs).

Statement of Procedure:

On June 7, 2000 XDS received three powder samples. These samples were received and logged in by XDS.  These samples were assigned WDS identification numbers (see appendix1). The samples were stored at approximately 4° C until they were extracted. The samples were extracted and cleaned up using our patent pending sample processing procedure.  

Just prior to dosing the H1.L1.6 mouse hepatoma cells, the sample extracts in DMSO were suspended in cell culture medium. This media was then used to expose monolayers of our cells grown in 96 well culture plates. In addition to the dilutions of samples, asStandard curve of TCDD concentrations was assayed TCDD (128.8, 64.4, 32.2, 16.1, 8.0, 4.0 2.0, 1.0, 0.5 and 0.2 parts per trillion (ppt) TCDD). All assays of standards and unknowns were run in duplicate. The plates were incubated for a time to induce optimal expression of luciferase activity in a humidified C02 incubator. Following the incubation the media was removed and the cells were microscopically observed for viability. The luciferase assay was run to quantify the induction of luciferase activity.

Summary of Results:

Cell viability: Microscopic examination of the cells following exposure to the sample extracts did not reveal any indication of toxicity to the cells.  

The samples were analyzed and compared to an XDS Method blank. All three samples were below 0.4pg/g. Mean TEQ determined in the sample is shown in Summary Table 1 as estimated from comparison of the response to 2,3,7,8-tetrachlorodibenzo-p-dioxin in our cell based system.
